EB1 is a highly conserved, important regulator of microtubule dynamics which stabilizes and stimulates growth at the plus-end of microtubules (Jayatilaka et al. 2018). The protein consists of two functional domains: the N-terminal CH domain recognizes the growing ends of microtubules, while the C-terminal EBH domain recruits microtubule plus-end tracking proteins (+TIPs) (van Haren et al. 2018) which also regulate microtubule dynamics and functions. Interaction between EB1 and +TIPs appears to depend on post-translational modifications of EB1. For example, phosphorylation of EB1, mediated by ASK1, contributes to the ability of EB1 to track plus-ends of microtubules (Ran et al. 2017). ASK1 promotes proliferation of pancreatic cancer cells, and EB1 phosphorylation has been found to mediate the function of ASK1 and contribute to pancreatic cancer development (Gao et al. 2017). EB1 overexpression has been identified in early stages of oral cancer (Kumar et al. 2016) and colorectal cancer (Stypula-Cyrus et al. 2014).
