    • Rabbit anti Human parkin (pSer101) antibody recognises the 465 amino acid ~52 kDa human parkin protein, when phosphorylated at Ser101. Rabbit anti Human parkin (pSer101) antibody does not recognize the non-phosphorylated protein.

      Parkin is an E3 ubiquitin ligase highly expressed in the brain, which functions in the ubiquitin-proteasome system, together with the E2 ubiquitin conjugating co-enzyme UbcH7. There are at least five phosphorylation sites in parkin including Ser101, shown to be phosphorylated by Casein kinase-1 (CK-1), and suggest that phosphorylation of parkin may act to regulate its ubiquitin ligase activity (Yamomoto et al. 2004). Mutations in the parkin gene are associated with the hereditary neurodegenerative disorder known as autosomal recessive early onset Parkinson disease 2 (PARK2), characterized by progressive dopaminergic neural cell death.

      Antisera to human parkin were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.

      For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
