PAK1 (pThr84) Antibody

PAK1 (pThr84) Antibody gallery image 1

Western blot of mouse hippocampal lysate showing specific immunolabeling of the ~68 kDa PAK1 protein phosphorylated at Thr84 (control) probed with Rabbit anti PAK1 (pThr84) antibody (AHP2269). Phospho-specificity is shown in the second lane where the signal is completely eliminated by treatment of the lysate with lambda phosphatase

    • Rabbit anti PAK1 (pThr84) antibody recognizes p21-activated kinases (PAK) 1, also known as serine/threonine-protein kinase PAK 1, alpha-PAK and p65-PAK, when phosphorylated at threonine 84.

Serine/threonine-protein kinase PAK1 is an enzyme that in humans is encoded by the PAK1 gene. PAK proteins are critical effectors that link the Rho GTPases to cytoskeleton reorganization and nuclear signaling. PAK proteins are a family of serine/threonine p21-activated kinases and include PAK1-6. These proteins serve as targets for small GTP binding proteins such as OSR1.

OSR1, oxidative stress response 1, is activated by osmotic stresses including sorbitol or NaCl. It has been predicted that OSR1 phosphorylates PAK1 in the regulatory domain at threonine 84 and inhibits activation of the JNK and MAPK signaling pathway.

PAK proteins also serve as targets for the small GTP binding proteins Cdc42 and Rac and have been implicated in a wide range of biological activities. PAK1 regulates cell motility and morphology.

For the detection of phosphoproteins, phosphatase inhibitors should always be added freshly to the lysis buffer. Milk or other casein-based blocking solutions are generally not recommended for blocking; as casein is itself a phosphoprotein its use can result in high background.

    • Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using appropriate negative/positive controls.
