    • Mouse anti Human GCH1 monoclonal antibody, clone 4A12 recognizes guanosine triphosphate cyclohydrolase 1, also known as GTP cyclohydrolase 1 or GTP-CH-I, encoded by the GCH1 gene. GTP cyclohydrolase 1 has multiple potential isoforms but only isoform 1 gives rise to an active form of the enzyme (Gütlich et al. 1994). GTP cyclohydrolase 1 has a predicted molecular weight of ~28kDa, possesses three zinc binding sites and has two potential phosphoserine residues (Widder et al. 2007) .

      GTP-CH 1 is the initial rate limiting factor involved in the biosynthesis of tetrahydrobiopterin (BH4), itself an essential co-factor in the generation of active forms of nitric oxide synthase and in catecholamine and 5-hydroxytryptamine (serotonin) biosynthesis (Thöny et al. 200).

      Deficiency of GTP-CH1 is implicated in malignant hyperphenylalaninemia due to tetrahydrobiopterin deficiency (Ichinose et al. 1995), whilst abnormalities of the GCH1 gene have also been documented for their involvement in locomotor pathologies including dystonia 5, a condition affecting gait and posture with marked fluctuating diurnal presentation (Hu et al. 2010).

      Mouse anti human GCH1 has been used effectively for the identification of GTP cyclohydrolase 1 expression in cells using Western blotting (Ryan et al. 2013).
