FOXO1 Antibody

FOXO1 Antibody gallery image 1

Western blot analysis of FOXO1 was performed by loading 25 ug of HeLa (lane 1), 293T (lane 2) and rat heart lysates (lane 3) onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked at 4°C overnight. The membrane was probed with FOXO1 polyclonal antibody at a dilution of 1/500 overnight at 4°C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Detection was performed using chemiluminescence. Images were taken at an exposure time of 2 min (lanes 1 and 2) and 1 min (lane 3). Results show a band at ~66 kDa

    • Rabbit anti FOXO1 antibody recognizes Forkhead box protein O1, also known as FOXO1, Forkhead box protein O1A or Forkhead in rhabdomyosarcoma.

FOXO1 like other members of the forkhead protein family acts as a transcription factor by binding to DNA with its forkhead box protein domain. The subcellular activation and thereby the transcriptional activity of FOXO1 is regulated via AKT mediated phosphorylation while its levels are regulated via polyubiquitination.

FOXO1 has been shown to play a role in various signaling pathways ranging from apoptosis, proliferation, oxidative stress, DNA damage to insulin signaling (Huang and Tindall 2007). The protein has also been proposed to act as a tumor suppressor (Fu and Tindall 2008, Xie et al..2012).
