Sandwich ELISA analysis of human ferritin using Mouse anti Human collagen I/II/III antibody, clone NLI/22 (2150-2203) as a capture reagent and biotinylated Mouse anti Human collagen I antibody, (131008) as a detection reagent with native human collagen (2150-0030) as antigen. Detection is by HRP conjugated streptavidin and substrate. Microtitre plate is read at O.D. 450 nm on the Bio-Rad iMark Microplate Absorbance Reader (Bio-Rad Catalog 168-1135). Normal serum (green) and plasma (orange) samples are included diluted 1:4.

Collagen is located in the extracellular matrix of connective tissues. It is part of the interacting network of proteoglycans and proteins that provides a structural framework for both soft and calcified connective tissues. Type I collagen (95 kDa) is found in bone, cornea, skin and tendon. Mutations in the encoding gene are associated with osteogenesis imperfecta, Ehlers Danlos syndrome, and idiopathic osteoporosis.

Preparation
Collagen from washed dissected tissue was dissolved in acetic acid after mild pepsin treatment. Collagen type I was then purified using differential salt precipitation.
