C) At 4d–5d post-injury, co-staining with anti-α-SMA (red) and anti-α-CAA (green) shows that during the healing process, only a few fibers co-express both isoforms after 4d (a–c, arrowhead), whereas after 5d, only α-CAA positive fibers are detected (d–f). After 4d, myofibroblasts might participate to the repair process and are detected by using the anti-α- SMA mAb (a–c, arrow). Merged images are shown on right column. Bars = 50μm.

Actins are highly conserved proteins that play a role in cell contractile activity, motility, cytokinesis, and in maintaining cell structure. Alpha smooth muscle actin is expressed in vascular smooth muscle, myofibroblasts and myoepithelial cells and also in tumours derived from these cells.

              Excellent for immunofluorescence in rat cells

              We previously had used the 1A4 clone from another major supplier who recently lost their license to sell antibodies. Looking for a replacement, we tried Bio-Rad's Actin Alpha Antibody 1A4 primary. We compared dilutions of both antibodies side-by-side and found equivalent staining of both, which appear to be highly specific for actin fibres in A7r5 rat aorta smooth muscle cells. We diluted the primary to 200 microg/ml. Using 4% normal goat serum as block, we get excellent staining at 300-500x dilution (0.4-0.67 microg/ml). One could probably go to 0.1-0.2 microg/ml if using a high NA objective and sensitive imaging system. We will post images soon.
