Detect DNA synthesis or apoptosis
Bromodeoxyuridine (BrdU) is a synthetic nucleoside and an analog of the DNA base thymidine. When administered to cells BrdU can replace thymidine during DNA synthesis. When BrdU is incorporated into the newly synthesized DNA of dividing cells it can be detected using anti-BrdU antibodies.

The use of antibody detection of incorporated BrdU into cellular DNA is an accurate method to measure cell proliferation both in vivo and in vitro, because BrdU can be easily diluted and administered into cultures, injection (Tesfaiqzi et al. 2014), or drinking water (Moser et al. 2004).
Extensively referenced for routine cell proliferation assays, clone BU1/75 (ICR1) has also been used in the detection of label retaining stem cells (Kimoto et al. 2008), label retaining neurons (Murata et al. 2011) and to study the speed of the DNA replication fork (Bugler et al. 2010).
Bio-Rad offers two popular anti-BrdU antibodies, a rat monoclonal BU1/75 (ICR1) and a mouse monoclonal Bu20a in formats suitable for flow cytometry, immunofluorescence and immunohistology-paraffin.
Bio-Rad also offers two kits for measuring the level of DNA fragmentation during late stage apoptosis. Both kits are called APO-BrdUTM TUNEL assay kits; one is specific for immunohistochemistry, and the other is used for flow cytometry.
The Apo-BrdU™ flow cytometry kit (APO001) is used for identifying cells during the late stages of apoptosis by flow cytometry and fluorescence microscopy. It can also be used in conjunction with cell surface staining, which should be performed first. The kit is a two color terminal deoxynucleotide transferase dUTP nickel end labeling (TUNEL) kit which labels DNA breaks, identifying fragmentation and total cellular DNA. This kit involves using a two color flow cytometer with green (515 nm) and red (635 nm) fluorescence detectors.
The Apo-BrdU™ immunohistochemistry kit (APO002) is ideal for detecting DNA breaks and fragmentation using immunohistochemistry detection on frozen (cryosections), paraffin embedded tissue sections and cytospins. This kit is also suitable for cell preparations fixed on slides.
