Human anti bevacizumab inhibition ELISA

A microtiter plate was coated over night with human VEGF at a concentration of 1 μg/ml. After washing and blocking with PBST+5% BSA, a pre-incubated mixture of bevacizumab (0.3 μg/ml) plus increasing concentrations of human anti bevacizumab (clone AbD17976, HCA182) in the monovalent Fab format was added. Free bevacizumab, still capable of binding to the human VEGF coated plate, was detected using a HRP-conjugated mouse anti human IgG (Fc) specific antibody (MCA647P) and QuantaBlu™ fluorogenic peroxidase substrate. Data is shown as the mean of three measurements

      • Bevacizumab, also known as Avastin, is a humanized monoclonal antibody (IgG1/kappa) that inhibits angiogenesis by binding to the vascular endothelial growth factor A (VEGF-A). VEGF-A stimulates new blood vessel formation, also in certain diseases, especially in cancer. Avastin is used to treat various cancers, including colorectal, lung, breast, glioblastoma, kidney and ovarian. Clinical trials are underway for many other indications.

        Clone AbD17976_hIgG1 is an anti-idiotypic antibody that specifically recognizes the Avastin humanised monoclonal antibody. This fully human antibody can be used to develop and calibrate immune response assays to measure the anti-drug antibody (ADA) response in patient sera.

        Human anti Bevacizumab antibody, clone AbD17976_hIgG1 is an affinity-matured antibody. The monovalent intrinsic affinity of HCA185 was measured as kDa = 0.4 nM by real time, label free molecular interaction analysis using an Attana A200 instrument on immobilized Avastin.
